UPCC 10914 Laboratory Analysis of Immune Cells in Patients with Malignancy
Brief description of study
The primary objective of this protocol is to collect and store peripheral blood samples from different acute and chronic, myeloid and lymphoid leukemias and non-leukemia cancer patients for research analysis. Freshly collected blood leukocytes will be separated from red blood cells on a ficoll gradient, counted, and stored in 5-10x10E6 aliquots. RNA will be extracted from one 5x106 aliquot. We will thaw and test 5-sample batches by flow cytometry for the cell-surface expression of previously described (for example CD33, CD123, CD34) and novel antigens (to be determined). Immune cells isolated from patient blood will be studied using multi-parameter flow cytometry +/- cell culture techniques and followed with functional assays (such as phagocytosis, killing, proliferation, and cytokine production) in the presence of autologous or allogeneic tumor cells.
