Mismatch Prospective Cohort Study (MPC)

Aim 1: To obtain measures of cerebral beta-amyloid plaques with amyloid PET (A), neurofibrillary tangles with tau PET (T), and neurodegeneration with structural MRI (N) in a cohort of older adults diagnosed with MCI or AD to evaluate this dataset with a T-N mismatch metricmodel for phenotypic discovery. The central conceptual goal of this proposal is to explicitly model the much richer, spatially varying T-N relationship than is currently conceived in the ATN framework, as a conduit to both help broaden the biomarker-based understanding of phenotypes, and also as a useful tool for cohort selection in therapeutic trials and eventually clinical practice.
